There are delays and diversions for motorists heading into Newcastle on the Link Road at Cameron Park.
Police say a red Holden Commodore failed to negotiate a roundabout at Minmi Road and hit a tree at about 12.30am on Thursday.
Police say a 29-year-old man was behind the wheel, while two men aged 20 and 21 were passengers along with a 24-year-old female.
All four people in the car were taken to hospital.
Police say one person suffered suspected spinal fractures and another suspected internal injures.
Police have set up a crime scene and the east bound lanes of the Newcastle Link Road are closed between Woodford Street and Minmi Road.
Traffic is being diverted via Cameron Park Drive, George Booth Drive and Lake Road to rejoin the Link Road.
